Malik lab at Cincinnati Children's Hospital Medical Center has led many gene therapy studies on hematopoietic stem cell disorders, such as hemoglobinopathies (thalassemia and sickle cell disease), immune deficiency (X-SCID) and immune dysregulatory disorders (HLH) and using a variety of viral vectors and have translated these efforts with retroviral and lentiviral vectors into clinical trials for these diseases. The lab is now focused on gene editing for hematopoietic diseases, with a focus on designing platforms that primarily mediate homology directed repair.
